Previous studies focused on cerebrospinal fluid (CSF) markers, which are imprecise for detecting subtle BBB disruption. Dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) enables sensitive investigation of subtle BBB leakage in vivo, yet remains unexplored in SSD research. We hypothesized higher leakage in SSDs compared to healthy controls (HCs), indicating a clinical sub-phenotype.Forty-one people with SSDs and forty age- and sex-matched HCs were included in this cross-sectional study employing DCE-MRI, clinical characterization, cognitive assessment, blood and CSF analyses. The volume transfer constant Ktrans, calculated using the Patlak method to estimate the contrast agent transfer between blood and extravascular space, was compared between groups to detect differences in BBB leakage.Individuals with SSDs showed higher BBB leakage compared to HCs in a widespread pattern, in brain regions typically affected in SSDs. No significant association was detected between leakage and measures of cognition, symptom severity, peripheral inflammation markers and albumin CSF/serum ratio.This is the first study to date reporting BBB leakage in SSDs compared to HCs in multiple brain regions implicated in the disorder. These findings provide insights into disease mechanisms, highlighting the need for further investigation into the role of the BBB in SSDs. 000278044 536__ $$0G:(DE-HGF)POF4-352$$a352 - Disease Mechanisms (POF4-352)$$cPOF4-352$$fPOF IV$$x0 000278044 588__ $$aDataset connected to CrossRef, PubMed, , Journals: pub.dzne.de 000278044 650_7 $$2Other$$aBBB 000278044 650_7 $$2Other$$aDCE-MRI 000278044 650_7 $$2Other$$aLeakage 000278044 650_7 $$2Other$$aNeuroimaging 000278044 650_7 $$2Other$$aSSD 000278044 650_7 $$2Other$$aSchizophrenia 000278044 7001_ $$aYakimov, Vladislav$$b1 000278044 7001_ $$aRoell, Lukas$$b2 000278044 7001_ $$0P:(DE-2719)9001808$$aRauchmann, Boris Stephan$$b3$$udzne 000278044 7001_ $$aToth, Hannah$$b4 000278044 7001_ $$aMelcher, Julian$$b5 000278044 7001_ $$aJäger, Iris$$b6 000278044 7001_ $$aLutz, Isabel$$b7 000278044 7001_ $$aKallweit, Marcel S$$b8 000278044 7001_ $$aPapazov, Boris$$b9 000278044 7001_ $$aBoudriot, Emanuel$$b10 000278044 7001_ $$aSeelos, Klaus$$b11 000278044 7001_ $$aDehsarvi, Amir$$b12 000278044 7001_ $$aCampana, Mattia$$b13 000278044 7001_ $$aRaabe, Florian$$b14 000278044 7001_ $$aMaurus, Isabel$$b15 000278044 7001_ $$aLöhrs, Lisa$$b16 000278044 7001_ $$0P:(DE-2719)9001539$$aBrendel, Matthias$$b17$$udzne 000278044 7001_ $$aStöcklein, Sophia$$b18 000278044 7001_ $$aFalkai, Peter$$b19 000278044 7001_ $$aHasan, Alkomiet$$b20 000278044 7001_ $$aGroup, Cdp Working$$b21 000278044 7001_ $$aFranzmeier, Nicolai$$b22 000278044 7001_ $$aKeeser, Daniel$$b23 000278044 7001_ $$aWagner, Elias$$b24 000278044 773__ $$0PERI:(DE-600)1462491-6$$a10.1016/j.bbi.2025.04.003$$gVol. 128, p. 256 - 265$$p256 - 265$$tBrain, behavior and immunity$$v128$$x0889-1591$$y2025 000278044 8564_ $$uhttps://pub.dzne.de/record/278044/files/DZNE-2025-00550.pdf$$yOpenAccess 000278044 8564_ $$uhttps://pub.dzne.de/record/278044/files/DZNE-2025-00550.pdf?subformat=pdfa$$xpdfa$$yOpenAccess 000278044 909CO $$ooai:pub.dzne.de:278044$$pdnbdelivery$$pdriver$$pVDB$$popen_access$$popenaire 000278044 9101_ $$0I:(DE-HGF)0$$6P:(DE-2719)9001808$$aExternal Institute$$b3$$kExtern 000278044 9101_ $$0I:(DE-588)1065079516$$6P:(DE-2719)9001539$$aDeutsches Zentrum für Neurodegenerative Erkrankungen$$b17$$kDZNE 000278044 9131_ $$0G:(DE-HGF)POF4-352$$1G:(DE-HGF)POF4-350$$2G:(DE-HGF)POF4-300$$3G:(DE-HGF)POF4$$4G:(DE-HGF)POF$$aDE-HGF$$bGesundheit$$lNeurodegenerative Diseases$$vDisease Mechanisms$$x0 000278044 9141_ $$y2025 000278044 915__ $$0StatID:(DE-HGF)0200$$2StatID$$aDBCoverage$$bSCOPUS$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0160$$2StatID$$aDBCoverage$$bEssential Science Indicators$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)1050$$2StatID$$aDBCoverage$$bBIOSIS Previews$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)1190$$2StatID$$aDBCoverage$$bBiological Abstracts$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0600$$2StatID$$aDBCoverage$$bEbsco Academic Search$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)9915$$2StatID$$aIF >= 15$$bBRAIN BEHAV IMMUN : 2022$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0100$$2StatID$$aJCR$$bBRAIN BEHAV IMMUN : 2022$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)1030$$2StatID$$aDBCoverage$$bCurrent Contents - Life Sciences$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0113$$2StatID$$aWoS$$bScience Citation Index Expanded$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0150$$2StatID$$aDBCoverage$$bWeb of Science Core Collection$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0510$$2StatID$$aOpenAccess 000278044 915__ $$0StatID:(DE-HGF)0030$$2StatID$$aPeer Review$$bASC$$d2024-12-19 000278044 915__ $$0StatID:(DE-HGF)0300$$2StatID$$aDBCoverage$$bMedline$$d2024-12-19 000278044 915__ $$0LIC:(DE-HGF)CCBY4$$2HGFVOC$$aCreative Commons Attribution CC BY 4.0 000278044 915__ $$0StatID:(DE-HGF)0420$$2StatID$$aNationallizenz$$d2024-12-19$$wger 000278044 915__ $$0StatID:(DE-HGF)0199$$2StatID$$aDBCoverage$$bClarivate Analytics Master Journal List$$d2024-12-19 000278044 9201_ $$0I:(DE-2719)1110007$$kAG Haass$$lMolecular Neurodegeneration$$x0 000278044 980__ $$ajournal 000278044 980__ $$aVDB 000278044 980__ $$aUNRESTRICTED 000278044 980__ $$aI:(DE-2719)1110007 000278044 9801_ $$aFullTexts
