Abstract: 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is a neurotoxicant that damages the nigrostriatal system and causes a parkinsonian motor syndrome in humans and experimental animal models. It is a classic protoxin because its fully oxidized 1-methyl-4-phenylpyridinium metabolite is actually responsible for neurotoxicity. Neuronal damage results from a cascade of deleterious events that include mitochondrial impairment. MPTP-based models, although not devoid of important limitations, have provided clues on pathogenetic processes relevant to Parkinson's disease (PD). Perhaps most importantly, they have been instrumental for the development of therapies against PD motor symptoms.
