Abstract: At excitatory synapses of the central nervous system, the postsynaptic density (PSD), a specialized structure underneath the postsynaptic membrane, controls the reception and integration of synaptic signals. The PSD functions as an extended protein assembly connecting integral membrane proteins (e.g., glutamate-gated ion channels) with cytoskeletal elements and signaling molecules via an ordered array of scaffolding proteins. Synaptic activity can adjust size and composition of the PSD, contributing to changes in synaptic strength.
