Abstract: Calcium Homeostasis Modulator Protein 6 or CALHM6 is a tetraspanin-like membrane glycoprotein from the family with sequence similarity 26, member F (FAM26F) that majorly contributes to regulating gene expression and various cellular processes. Substantial evidence supports CALHM6 to be a calcium modulator with a potential immune-modulatory role in various infections and cancer, however, the associated signaling cascades and related functional details are largely unknown and have yet to be investigated for its exploitation as a therapeutic candidate. The current study exploited mass spectrometry-based proteome analysis to analyze differentially expressed proteins in in-vitro knockdown (via a siRNA-based approach ) as well as overexpression (plasmid-mediated) models of CALHM6. Overexpression and knockdown were confirmed at the mRNA level using real-time PCR and at the protein level using western blotting. Cell lysates subjected to proteome analysis 24 h post-treatment and mass spectrometry analysis was performed. Total of 5282 proteins were found in desired comparison groups i.e. Overexpression (OE) vs. siRNA control (SCR), Knockdown (KD) vs. SCR, and OE vs. KD. Python analysis and pathway enrichment was performed using Metascape.Current study reveals valuable insights into the pathways and proteins identified during overexpression and knockdown of CALHM6, shedding light on the complex molecular mechanisms governing the immunomodulatory functions of CALHM6.
