Abstract: During development in the central nervous system oligodendrocytes produce large amount of tightly compacted myelin membranes around the axons. Myelin turnover is crucial for the maintenance of functional nerves. However, it is not known how myelin molecules get access to the lysosomal degradation system for turnover of myelin membrane. We found that in the aging brain, myelin fragments are formed by shedding myelin sheaths from the axon into the extracellular space and subsequently engulfed by microglia. Internalized myelin fragments are associated with insoluble lysosomal inclusions known as lipofuscin within microglia. Then we suggest that upon increase of myelin fragmentation with age, microglial lysosomal system become overloaded, contributing to lysosomal storage and subsequently aging phenotype.
