Abstract: Due to the regulatory function of the human blood-brain barrier (BBB), optimizing brain-targeting drugs for brain-penetration is a challenge in drug discovery. Therefore, the development of in vitro assays that mimic relevant properties of the BBB are in focus to better predict drug permeability in humans. This article describes human iPSC-derived brain endothelial microvessels in perfusable multi-chip plates that provide physiological properties of the BBB in a scalable format for screening.
