Abstract: Tau is a microtubule-associated protein found mainly in the axons of neurons in the brain. Abnormal changes in Tau (e.g., aggregation, hyperphosphorylation) are hallmarks of Alzheimer's disease. Two processes of relocalization of Tau may be related to early states of the pathology and have received much attention: (1) the redistribution of Tau within cells (termed 'somatodendritic missorting') and (2) the release and reuptake of Tau from donor to acceptor cells (termed 'spreading'). Because of the tripartite nature of neurons (cell body, dendrites, axons), these changes can be studied by microfluidic chambers (MFCs) which allow separation and observation of Tau in neuronal compartments. In this chapter, we present some methods and research results obtained by using microfluidic devices.
